A lead compound containing a methyl ester functional group was hydrolysed to give an analogue with a carboxylic acid functional group. An in vivo bioassay which involved oral administration suggested that the ester was active and that the acid was inactive. However, an in vitro bioassay suggested that the ester was inactive and that the acid was active.
